Application of Transgenic Technology in Apple Breeding

1 Breeding pest-resistant varieties At present, insect-resistant transgenic plants have been obtained on apples. The main insect-resistant genes used are Bt (Bacillus thypsin inhibitor) and CpTI genes. The Bt gene is an insecticidal crystal protein (ICP) gene isolated from Bacillus thuringiensis. As early as 1991, the ICP gene was successfully used to transform apples in the United States. In 1992, the United States approved that it would carry field tests of transgenic apple plants with ICP genes. Domestic Cheng Jiasheng et al. also used Bt genes to successfully obtain transgenic apple plants and carried out insect-resistant tests. CpT I is resistant to many pests, and its broad spectrum is its main advantage. The use of CpT I gene in foreign countries in 1992 successfully transformed the green-sleeved apple; domestic Ducton and other CpT I genes were also used to transform the Royal Gala, Southem hybridization proved that the target gene exists in the apple genome.
Apple disease-resistant transgenic research focused on the resistance to fire blight (Erwina amylovoea), the main application of attacinE, cecropinSB-37, cecropm Shiva-1 genes. Norelli et al. used the four genes encoding lysed bacterial cells (attacinE, SB-37, Shiva-1, and hen white lysozyme) to obtain 250 transformation systems using Agrobacterium-mediated transformation of rootstock M7 and Royal Jura, identified in the greenhouse and in the field. The transformed plants carrying attacinE showed great resistance to the disease; the attaciton E gene was used to transform M7, and the transformed plants obtained had greatly increased resistance to fire blight, and some even reduced the disease by 50%. It has also been reported that Shiva-1 and attacin E are introduced into the Royal Gala, but transgenic plants tested in greenhouses are not stable against fire blight.
2 Dwarfing varieties have been identified and cloned from Agrobacterium tumefaciens. Some genes related to the synthesis and metabolism of plant hormones, mainly ipt genes and rolA, rolB, rolC, rolD genes, etc. In vivo expression may exhibit dwarfing traits in plants. Trifonova et al used Agrobacterium-mediated method to introduce ipt gene into Granny Smith, and the obtained transgenic plants showed normal appearance except for two strains that showed “medium irrigation”. Holefors et al. used rolA gene to transform rootstock M26, and obtained transgenic plants with short trees, among which three lines were significantly shortened, and three lines had lower leaf and root dry weight. In order to increase dwarfing ability of semi-dwarf rootstock M26, rolA, rolABC, Arabidopsis phyB and oat phyA genes were introduced into M26. As a result, it was found that the first three genes were overexpressed in the transgenic plants and the trees showed different degrees of Dwarfing ability.
3 Breeding of easy-to-root varieties is controlled by genes. Some apple rootstocks are extremely difficult to root when cutting and layering. In order to increase the rooting ability of M26 rootstock, Lamber et al. imported the T-DNA region from Ri plasmid of A. rhizogenes into M26. The transformed plants carrying the T-DNA of the Ri plasmid had good root systems and greatly improved rooting ability. The roIB gene induces hair root formation on many host plants. Welander et al. used roIB gene to study the transformation of rootstock M26. Transgenic plant roots were more sensitive to auxin and the rooting capacity was also increased.
4 Selection of cultivars for cultivars Apples are difficult to control due to fruit ripening during storage, which often leads to overcooking and rot, resulting in great economic losses. In recent years, with the gradual deepening of molecular biology research, it has become possible to use genetic engineering techniques to improve the storage of apples. The ACC oxidase gene, ACC synthase gene, and PG (multiple galactose synthase) gene have been cloned from various plants. On apples, the use of antisense ACC oxidase, antisense ACC synthase, and antisense PG genes to transform royal jelly apples to improve storage stability is underway. Transgenic plants have been obtained and it is expected that the ideal resistance will be developed soon. Storage species.
5. Herbicide-tolerant herbicide-tolerant genetic engineering has been successful in the selection of herbicide-tolerant herbicides. At present, als genes are commonly used in plant genetic transformation. In apples, the Arabidopsis als gene was transformed into Royal Gala apple by Agrobacterium-mediated transformation to obtain transgenic plants. In subsequent studies, the seeds obtained were tested for resistance with a 60 mg/L Glean (green barley) spray and it was found that the als gene was stably inherited in a 1:1 ratio.
